A blood-brain barrier-permeant probe for live amyloid plaques imaging by fluorescent 2-photon microscopy.
Methoxy-X04 (MeO-X04, MethoxyX04, MeX04, MX04) is a blood-brain barrier-permeant fluorescent probe derived from the Chrysamine G and Congo red scaffold that retains affinity toward amyloid beta (Aβ) fibrils (Ki = 25.3 nM/Chrysamine G and 26.8 nM/Methoxy-X04), while being much more lipophilic than Chrysamine G and Congo red. Methoxy-X04 stains Aβ plaques, tangles, and cerebrovascular amyloid in postmortem brain sections with good specificity and is commonly utilized for high-resolution fluorescent imaging of individual amyloid plaques in live animals by 2-photon microscopy (Ex. 750-800 nm; Em. 380-480, 500-540, and 560-650 nm) following i.v. or i.p. injection (5-10 mg/kg).
