General description
Amphoterin-induced protein 2 (UniProt: Q86SJ2; also known as AMIGO-2, Alivin-1, Differentially expressed in gastric adenocarcinomas (DEGA)) is encoded by the AMIGO2 (also known as ALI1) gene (Gene ID: 347902) in human. AMIGO-2 is a member of the AMIGO family, which includes three, type I transmembrane proteins: AMIGO-1, AMIGO-2, and AMIGO-3. These proteins are known to play a role in nervous system development and have been implicated in various neuronal activities. AMIGO-2 protein contains 6 leucine-rich repeats (LRR), an Ig-like C2-type domain in the extracellular region, a transmembrane domain (aa 399-419), and a cytoplasmic domain (aa 420-522). It is involved in cell adhesion, axon tract development, and synaptic formation within the nervous system. The AMIGO family of proteins exhibit both homophilic and heterophilic binding activity. AMIGO-2 is primarily expressed in the nervous system. Its expression is typically higher during the early stages of brain development. AMIGO-2 have been linked to several neurological, neurodevelopmental disorders and can influence neural regeneration processes. Changes in AMIGO-2 expression levels have been observed in certain cancers and it is differentially expressed in human gastric adenocarcinomas. Application
Quality Control Testing
Evaluated by Western Blotting in K562 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected AMIGO-2 in K562 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected AMIGO-2 in lysates from Y79, A549 cells and human kidney tissue.
Affinity Binding Assay: A representative lot of this antibody bound AMIGO-2 peptide with at least one hundred thousand-fold (100,000X) higher than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected AMIGO-2 in K562 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected AMIGO-2 in human stomach tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1B9 is a ZooMAb® rabbit recombinant monoclonal antibody that detects AMIGO-2. It targets an epitope within 26 amino acids from the C-terminal extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
