m-Diazonium BODIPY is a fluorogenic probe engineered for site-specific labeling of tyrosine residues in amino acids, peptides, proteins, and intricate biomolecules via an azo-coupling reaction, facilitating the formation of fluorescent biosensors. The design of BODIPY m-diazonium ensures precise targeting and uniform labeling under physiological conditions, achieving high conversion rates during the coupling process without inducing epimerization of the asymmetric carbon in L-Tyr-OH. Tyrosine-specific BODIPY labeling of FKBP12 enables selective fluorescence detection of tacrolimus (KD = 45 nM) with a sensitivity threshold of 190 nM. The excitation and emission wavelengths are approximately 495 nm and 525 nm, respectively.
