Selective transient receptor potential cation channel TRPV4 blocker in cultures and in vivo.
GSK205 is a selective transient receptor potential cation channel TRPV4 blocker that prevents hyposmotic stress-, as well as TRPV4 activator 4αPDD-induced Ca2+ response (IC50 = 600 nM against 10 µM 4αPDD (EC50 = 2 µM); porcine articular chondrocytes). GSK205 upregulates the expression of thermogenic genes in 3T3-F442A adipocyte cultures (5 µM for 4 days) and in diet-induced obese (DIO) mice in vivo (10 mg/kg b.i.d. i.p.)
