Cancer tissue-targeting photosensitizer that induces irradiation-dependent cancer killing and in vivo photodynamic therapy (PDT) efficacy.
Talaporfin is a chlorin based cancer tissue-targeting photosensitizer that, upon irradiation, generates reactive oxygen species (ROS) that cause DNA damage and apoptosis in cancer cells. Talaporfin induces irradiation-dependent cancer killing in cultures (24 h 10-100 μg/mL Talaporfin sodium followed by washing and 664 nm laser irradation at 15 mW/cm2 and 10 J/cm2; ESCC) and displays photodynamic therapy (PDT) efficacy in human cancer exnograft models in mice in vivo, including EMT-6 sarcoma (100% cure with 664 nm laser at 100 J/cm2 and 100 mw/cm2 24h after 8 mg/kg talaporfin i.p.) and ESCC (664 nm, 100 J/cm2 2h after 10 mg/kg talaporfin sodium via tail vein i.v.).
