General description
Type-1 angiotensin II receptor (UniProt: P30556; also known as AT1AR, AT1BR, Angiotensin II type-1 receptor, AT1 receptor) is encoded by the AGTR1 (also known as AGTR1A, AGTR1B, AT2R1, AT2R1B) gene (Gene ID: 185) in human. AGTR1 is a multi-pass membrane glycoprotein of the G-protein coupled receptor family with seven transmembrane domains. It serves as a receptor for angiotensin II that acts as a key regulator of blood pressure and sodium retention by the kidney. The activated receptor couples to G- protein G(q) and thus activates phospholipase C and increases the cytosolic Ca2+ levels, which in turn triggers cellular responses such as stimulation of protein kinase C. Over-stimulation of ATGR1 leads to hypertension, cardiovascular hypertrophy, and fibrosis, whereas blocking its activity reduces blood pressure. Application
Quality Control Testing
Evaluated by Western Blotting in Human liver tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected AGTR1 in Human liver tissue lysate.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound AGTR1 peptide with a KD of 4.0 x 10-6 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 Dilution of this antibody detected AGTR1 in HeLa, HepG2, and NIH3T3 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1I12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Type-1 angiotensin II receptor (AGTR1). It targets an epitope within 14 amino acids from the first extracellular domain in the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
