General description
Non-receptor tyrosine-protein kinase TYK2 (UniProt: P29597; also known as Tyk2) is encoded by the Tyk2 gene (Gene ID: 7297) in human.TYK2 is a ubiquitously expressed member of the Janus kinase (JAK) family that is associated with receptors of type I interferon, interleukins 6, 10, 12, and 23 and plays a key role in the signal transduction of these cytokines. It is important in both innate and adaptive immune cells and is shown to be an essential component of maintaining normal immune responses. TYK2 is also involved in various other processes, including cell growth, development, and cell migration. It is reported to associate with growth factor receptors and activates STAT family of transcription factors. STAT proteins that homo- or heterodimerize following their phosphorylation translocate to the nucleus and regulate cytokine responsive genes. Its protein kinase 1 domain (pseudokinase domain), located in amino acids 589-875, mediates autoinhibition of the TYK2 kinase domain. Phosphorylation of tyrosine 1054 and 1055 induces kinase domain (aa 897-1176) activation. Application
Quality Control Testing
Evaluated by Western Blotting in Raji cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TYK2 in Raji cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TYK2 in Jurkat and COS-7 cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound TYK2 peptide with a KD of 8.7 x 10-9 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TYK2 in Human fallopian tube tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2H22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Non-receptor tyrosine-protein kinase TYK2. It targets an epitope within 23 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
