General description
Double-strand break repair protein MRE11 (UniProt: P49959; EC:3.1.-.-; also known as Meiotic recombination 11 homolog 1, MRE11 homolog 1, Meiotic recombination 11 homolog A (MRE11 homolog A)) is encoded by the MRE11 (also known as HNGS1, MRE11A) gene (Gene ID: 4361) in human. The double-strand break repair protein MRE11 is part of the MRN complex, which also includes RAD50 and NBS1. MRE11 itself is a nuclease that features both exonuclease and endonuclease activities. It has a conserved nuclease domain and is structured to form homodimers, a formation that is crucial for its activity in DNA repair. MRE11 plays a critical role in the detection, signaling, and repair of DNA double-strand breaks (DSBs), a type of DNA damage that can lead to cell death or cancer if not correctly repaired. MRE11′s nuclease activity helps process DSBs to create single-stranded DNA tails, which are essential for the subsequent steps of homologous recombination repair. Additionally, the MRN complex, with MRE11 at its core, is involved in activating the ATM kinase, a key player in the DNA damage response that coordinates cell cycle checkpoints and repair. MRE11 is ubiquitously expressed in all cell types, given its fundamental role in maintaining genomic stability. Its expression is tightly regulated according to the cell cycle to meet the needs for DNA repair at different stages of cell division. Mutations or dysregulation of MRE11 can lead to various diseases, particularly cancer. Deficiencies in MRE11 function impair the DNA damage response and can lead to genomic instability, a hallmark of cancer cells. Additionally, MRE11 mutations are associated with Ataxia telangiectasia-like disorder, a rare autosomal recessive disorder characterized by neurodegeneration, immunodeficiency, and heightened sensitivity to ionizing radiation. This condition underscores the importance of MRE11 in neurodevelopment and immune system function.

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human MRE11.
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected MRE11 in HeLa cell lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:1000 dilution from a representative lot detected MRE11 in HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound MRE11 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.
Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated MRE11 in HeLa cell lysate.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1E7 is a ZooMAb® recombinant rabbit monoclonal antibody that detects MRE11. It targets an epitope within 15 amino acids from the C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
