General description
Proliferating cell nuclear antigen (UniProt: P12004; alsoknown as PCNA) is encoded by the PCN gene (Gene ID: 5111) in human. PCNA is a homotrimeric nuclear protein thatserves as an auxiliary protein of DNA polymerase delta and is involved in thecontrol of eukaryotic DNA replication by increasing the polymerase′sprocessability during elongation of the leading strand. It Induces a robuststimulatory effect on the 3′-5′ exonuclease and 3′-phosphodiesterase, but notapurinic-apyrimidinic (AP) endonuclease, APEX2 activities. It is reported tocolocalize with CREBBP, EP300 and POLD1 to sites of DNA damage. It acts as aloading platform to recruit DDR proteins that allow completion of DNAreplication after DNA damage and promote post-replication repair. PCNA canundergo phosphorylation on tyrosine 211 by EGFR that stabilizeschromatin-associated PCNA. Following DNA damage, it can undergomonoubiquitination to stimulate direct bypass of DNA lesions by specialized DNApolymerases or polyubiquitination to promote recombination-dependent DNAsynthesis across DNA lesions by template switching mechanisms. Mutation in PCN gene are known to causeAtaxia-telangiectasia-like disorder 2 (ATLD2) that is characterized bydevelopmental delay, ataxia, hearing loss, and short stature. Immunogen
GST/His-tagged recombinant fragment corresponding to the first 150 amino acids from the N-terminal region of human Proliferating cell nuclear antigen (PCNA).
Application
Anti-PCNA, clone 3B22 ZooMAb, Cat. No. ZRB1442, is a recombinant Rabbit monoclonal antibody that specifically targets PCNA and is tested for in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PCNA in HeLa cell lysate.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PCNA in HeLa, A431, HUVEC cell lines.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PCNA in human tonsil tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound recombinant PCNA with a KD of 2.0 x 10-7 in an affinity binding assay.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Clone 3B22 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Proliferating cell nuclear antigen (PCNA). It targets an epitope within the first 150 amino acids.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in lyophilized from PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
