General description
Essential MCU regulator, mitochondrial (UniProt: Q9H4I9; also known as Single-pass membrane protein with aspartate-rich tail 1, mitochondrial ) is encoded by the SMDT1 (also known as C22orf32, EMRE) gene (Gene ID: 91689) in human. SMDT1 is a critical component of the mitochondrial calcium uniporter (MCU) complex, located in the mitochondrial inner membrane. This single-pass membrane protein plays a pivotal role in regulating calcium uptake into mitochondria, which is vital for cellular bioenergetics, signaling, and survival. SMDT1 bridges the calcium-sensing proteins MICU1 and MICU2 with the calcium-conducting MCU subunit, forming the uniporter complex. This interaction facilitates calcium transport into the mitochondrial matrix, directly influencing ATP production via oxidative phosphorylation and buffering cytosolic calcium levels during stress. The gene is ubiquitously expressed across tissues, ensuring proper mitochondrial calcium homeostasis. Dysregulation or variants of SMDT1 lead to impaired mitochondrial calcium uptake, often resulting in abnormal mitochondrial dynamics and muscle-related disorders, such as Spastic Ataxia 5, and are implicated in pancreatic ductal adenocarcinoma (PDAC) prognosis. Current research highlights SMDT1 as a therapeutic target, with potential applications in restoring mitochondrial function in conditions caused by calcium homeostasis disruption. KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal region of human SMDT1.
Application
Quality Control Testing
Evaluated by Western Blotting in COS-7 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SMDT1 in COS-7 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SMDT1 in human brain tissue lysate and HEK-293 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with SMDT1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 Dilution of this antibody detected SMDT1 in HEK-293 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2F16 is a ZooMAb® rabbit recombinant monoclonal antibody that detects SMDT1. It targets an epitope within 20 amino acids from the C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
