General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Oxidized low-density lipoprotein receptor 1 (UniProt: P78380; also known as Ox-LDL receptor 1, C-type lectin domain family 8 member A, Lectin-like oxidized LDL receptor 1 (LOX-1; Lectin-like oxLDL receptor 1; hLOX-1), Lectin-type oxidized LDL receptor 1, Oxidized low-density lipoprotein receptor 1, soluble form) is encoded by the OLR1 (also known as CLEC8A, LOX1) gene (Gene ID: 4973) in human. Oxidized low-density lipoprotein receptor 1 (OLR1), also known as LOX-1, is a transmembrane protein belonging to the C-type lectin superfamily. Structurally, it consists of a short intracellular domain, a transmembrane region, and an extracellular lectin-like domain that facilitates binding to modified lipoproteins, such as oxidized LDL (oxLDL). This receptor plays a critical role in vascular biology by mediating the recognition, internalization, and degradation of oxLDL, which is central to its function as a scavenger receptor. OLR1 is localized to the plasma membrane, nucleoplasm, and vesicles, indicating its dynamic role in cellular processes. OLR1 is involved in inflammation, oxidative stress, and apoptosis. It regulates pathways such as nuclear factor- B (NF- B) signaling, which drives the expression of pro-inflammatory cytokines and adhesion molecules. OLR1 expression is typically low under physiological conditions but is upregulated in response to atherosclerotic stimuli, including tumor necrosis factor-alpha (TNF- ), shear stress, and oxLDL exposure. This upregulation enhances endothelial dysfunction and promotes atherogenesis. OLR1 also plays roles in other pathological contexts, including ischemic stroke, diabetes, and cancer, where its activity contributes to oxidative stress and inflammation. OLR1 interacts with various proteins and signaling pathways, including those involved in lipid metabolism, immune cell activation, and apoptosis. Its expression is regulated by cyclic AMP signaling and is influenced by inflammatory mediators. Mutations in the OLR1 gene have been associated with increased susceptibility to myocardial infarction and other cardiovascular diseases. Soluble OLR1 (sOLR1), which is derived from proteolytic cleavage of the membrane-bound receptor, serves as a biomarker for vascular inflammation and atherosclerosis. Immunogen
KLH-conjugated linear peptide corresponding to 16 amino acids from the extracellular domain of human OLR1.
Application
Quality Control Testing
Evaluated by Western Blotting in RAW 264.7 cell lysate.
Western Blotting Analysis: 1:1,000 dilution of this antibody detected OLR1 in RAW 264.7 cell lysate.
Tested Applications
Western Blotting Analysis: 1:1,000 dilution from a representative lot detected OLR1 in human, mouse, and rat kidney tissue lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected OLR1 in human, rat, and mouse kidney tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound with OLR1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2E2 is a ZooMAb® rabbit recombinant monoclonal antibody that detects OLR1. It targets an epitope within 16 amino acids from the extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
