General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Charged multivesicular body protein 4b (UniProt: Q9H444; also known as Chromatin-modifying protein 4b (CHMP4b), SNF7 homolog associated with Alix 1, SNF7-2 (hSnf7-2), Vacuolar protein sorting-associated protein 32-2 (Vps32-2; hVps32-2)) is encoded by the CHMP4B (also known as C20orf178, SHAX1) gene (Gene ID: 128866) in human. The charged multivesicular body protein 4B (CHMP4B) is a core component of the Endosomal Sorting Complex Required for Transport III (ESCRT-III), which plays a critical role in membrane remodeling and scission events. Structurally, CHMP4B forms filamentous assemblies on membranes, promoting negative curvature and facilitating processes such as multivesicular body (MVB) formation, cytokinesis, and viral budding. These filaments are assembled into circular arrays that stabilize outward budding and scission of membranes. CHMP4B interacts with other ESCRT proteins, such as ALIX and CC2D1A, forming complexes that regulate its activity during vesicle trafficking and membrane repair. CHMP4B is ubiquitously expressed across human tissues, with significant roles in cellular homeostasis. It is involved in pathways regulating the degradation of cell surface receptors, exosome biogenesis, and autophagy. Dysregulation of CHMP4B has been implicated in pathological processes, including neuronal apoptosis during intracerebral hemorrhage (ICH) and potential contributions to neurodegenerative diseases. Its interaction with ALIX, a mediator of vesicle trafficking, further highlights its importance in cellular stress responses. In the context of disease, CHMP4B overexpression or malfunction has been associated with disruptions in membrane repair and cellular signaling, leading to apoptosis and inflammatory responses. Experimental studies suggest its role in stabilizing membrane curvature, which is crucial for maintaining organelle integrity and preventing pathological membrane ruptures. Immunogen
KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal half of human CHMP4B.
Application
Quality Control Testing
Evaluated by Western Blotting in HT-29 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CHMP4B in HT-29 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CHMP4B in MCF7, C2C12, and PC-12 cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound with CHMP4B peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CHMP4B in human small intestine and human cerebral cortex tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2H3 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CHMP4B. It targets an epitope within 16 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
