General description
Peroxisomal membrane protein PEX14 (UniProt: O75381; also known as PTS1 receptor-docking protein, Peroxin-14, Peroxisomal membrane anchor protein PEX14) is encoded by the PEX14 gene (Gene ID: 5195) in human. PEX14 is a single-pass membrane protein with a peroxisomal matrix domain (aa 2-108), a transmembrane domain (aa 109-126), and a cytosolic domain (aa 127-277). PEX14 is a multi-tasking protein that facilitates peroxisomal protein import and is also required for peroxisome motility by serving as membrane anchor for microtubules. PEX14 is integrated into peroxisome membranes with its C-terminus exposed to the cytosol and interacts with the cytosolic receptor for proteins containing a PTS1 peroxisomal targeting signal. It interacts with PEX13 to form the PEX13-PEX14 docking complex and is an essential component of the peroxisomal translocon that mediates import of cargo enzymes into peroxisomes. It also interacts with PEX5 via WxxxF/Y motifs. Mechanistically, PEX5 receptor along with cargo proteins associates with the PEX14 subunit of the PEX13-PEX14 docking complex in the cytosol, leading to the insertion of the receptor into the organelle membrane with the concomitant translocation of the cargo into the peroxisome matrix. Two isoforms of PEX14 have been described that are produced by alternative splicing. Application
Quality Control Testing
Evaluated by Western Blotting in HepG2 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PEX14 in HepG2 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PEX14 in lysates from NIH3T3 and C2C12 cells and Human Small intestine tissue and 1:1,000 dilution detected PEX14 in MDA-MB-23 cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PEX14 in Human small intestine tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PEX14 in HepG2 cells.
Flow Cytometry Analysis: 1 µg from a representative lot detected PEX14 in one million HepG2 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1D7 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detect Peroxisomal membrane protein PEX14. It targets an epitope within 23 amino acids from the C-terminal, cytoplasmic domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
