General description
Endoplasmic reticulum resident protein 29 (UniProt: P30040; also known as ERp29, Endoplasmic reticulum resident protein 28, ERp28, Endoplasmic reticulum resident protein 31, ERp31) is encoded by the ERP29 (also known as C12orf8, ERP28) gene (Gene ID: 10961) in human. ERp29 is ubiquitously expressed, homodimeric protein that resides in the lumen of endoplasmic reticulum (ER). It is mostly expressed in secretory tissues and plays an important role in the processing of secretory proteins within the ER, possibly by participating in the folding of proteins in the ER. ERp29 promotes ER to cis-Golgi cargo protein transport via COP II machinery through its interactions with the KDEL receptor; this interaction may facilitate the loading of ERp29 clients into COP II vesicles. It contains four distinct domains: An N-terminal ER localization domain (aa 1-32) that is cleaved upon entry into the ER; the b-type PDI domain, a highly conserved D-domain, and an ER retrieval KEEL motif (aa 258-261) at the C-terminus that prevents its secretion from ER. It lacks a classical thioredoxin (C-X-X-C) motif and is therefore devoid of thioredoxin enzymatic activity. ERp29 expression is reported to be upregulated during ER stress and the unfolded protein response (UPR) in pancreatic β-cells and thyroid epithelial cells. Two isoforms of ERp29 have been described that are produced by alternative splicing. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ERp29 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ERp29 in A431 and HepG2 cell lysates.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ERp29 in HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound full-length recombinant Human ERp29 with a KD of 2.1 x 10-8 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected ERp29 in Human kidney tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1H8 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Endoplasmic reticulum resident protein 29 (ERp29).
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
