General description
Nucleotide triphosphate diphosphatase NUDT15 (UniProt: Q9NV35; also known as EC:3.6.1.9, MutT homolog 2, MTH2, Nucleoside diphosphate-linked moiety X motif 15, Nudix motif 15, Nucleoside diphosphate-linked to another moiety X hydrolase 15, Nudix hydrolase 15) is encoded by the NUDT15 (also known as MTH2) gene (Gene ID: 55270) in human. NUDT5 is a homodimeric protein that catalyzes the hydrolysis of nucleoside triphosphates to their oxidized forms like 8-oxo-dGTP and the prodrug thiopurine derivatives 6-thio-dGTP and 6-thio-GTP. It converts 8-oxo-dGTP and 8-oxo-dGDP to 8-oxo-dGMP. However, it has considerably lower activity towards oxidized guanine species than the undamaged nucleotides. It exhibits stronger activity towards 2′-Deoxyguanosine-5′-triphosphate (dGTP).Its Nudix hydrolase domain is localized in amino acids 9-145 and its Nudix box motif resides in amino acids 48-69, which contains residues responsible for magnesium ion binding. Although NUDT15 exhibits high degree of homology with NUDT1/MTH1, but unlike MTH1 it is not active towards adenosine nucleotides, and they have different residues that make up the base of their active sites that confers different substrate recognition. NUDT15 plays a role in DNA synthesis and cell cycle progression by stabilizing PCNA. Its PCNA binding site is located in amino acids 76-164. Knockdown of NUDT15 is shown to promote PCNA degradation and UV irradiation is reported to accelerate PCNA degradation by inducing dissociation of PCNA-NUDT15. Application
Quality Control Testing
Evaluated by Western Blotting in HT-29 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected NUDT15/MTH2 in HT-29 cell lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected NUDT15/MTH2 in HT-29 cells.
Affinity Binding Assay: A representative lot of this antibody bound recombinant Human NUDT15/MTH2 protein with a KD of 2.6 x 10-7 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected NUDT15/MTH2 in Human liver tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1M22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects NUDT15/MTH2.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
