General description
Integrin alpha-L (UniProt: P20701; also known as CD11 antigen-like family member A, Leukocyte adhesion glycoprotein LFA-1 alpha chain, LFA-1A, Leukocyte function-associated molecule 1 alpha chain, CD11a) is encoded by the ITGAL (also known as CD11A) gene (Gene ID: 3683) in human. Integrins are heterodimeric, integral membrane proteins composed of a variable -subunit and a conserved β-subunit that function in cell surface adhesion and signaling. They contain a large extracellular domain responsible for ligand binding, a single transmembrane domain, and a cytoplasmic domain. The exact combination of various and β-subunits dictates the binding specificity of integrins to different ECM components. Although both subunits are required for adhesion, the binding specificity primarily depends on the extracellular portion of the -subunit. Application
Quality Control Testing
Evaluated by Western Blotting in THP-1 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CD11a in THP-1 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CD11a in lysates from Jurkat cells and Human spleen tissue.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD11a in Human spleen tissue sections.
Flow Cytometry Analysis: 0.1 µg from a representative lot detected CD11a in one million Human peripheral blood mononuclear cells (PBMC)..
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD11a in MV4-11 cells.
Affinity Binding Assay: A representative lot of this antibody bound in CD11a peptide with a KD of 2.9 x 10-7 in an affinity binding assay.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2J15 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects CD11a. It targets an epitope within 23 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
