General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Paired box protein Pax-8 (UniProt: Q06710; also known as PAX-8) is encoded by the Pax8 gene (Gene ID: 7849) in human. PAX-8 is a member of the paired box (PAX) family of transcription factors. Members of this gene family typically encode proteins which contain a paired box domain, an octapeptide, and a paired-type homeodomain. PAX proteins are important regulators of organogenesis and act as key factors in maintaining pluripotency of stem cells during development. PAX-8 is a nuclear protein that acts as a transcription factor for the thyroid-specific expression of the genes and is exclusively expressed in the thyroid cell type, maintaining the functional differentiation of such cells. PAX-8 is shown to be crucial in determining cell fate during the development of the thyroid, kidney, brain, eyes, and Müllerian system. It also regulates the expression of the Wilms′ tumor suppressor gene (WT1). PAX-8 is a shown to be a highly sensitive marker for thyroid, renal, Müllerian, and thymic tumors. However, lung adenocarcinomas, breast and adrenal neoplasms, and the majority of gastrointestinal tumors are reported to be negative for PAX-8. Mutations in Pax8 gene are known to cause congenital hypothyroidism that is characterized by thyroid dysgenesis and in some cases the thyroid gland can be completely absent, ectopically located and/or severely hypoplastic. Five isoforms of PAX-8 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Immunogen
KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal region of human PAX8.
Application
Quality Control Testing
Evaluated by Immunocytochemistry in HeLa cells.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected PAX8 in HeLa cells.
Tested Applications
Enzyme Immunoassay: A representative lot of this antibody detected PAX8 in ELISA application.
Affinity Binding Assay: A representative lot of this antibody bound PAX8 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 3N17 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PAX8. It targets an epitope within 16 amino acids from the C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
