MAO-N3, a cell-permeant HIDE (High-Density Environmentally-sensitive) probe, is tailored for precise targeting of the inner mitochondrial membrane, and when combined with SiR-DBCO, it forms the MAO-SiR complex, the anticipated product of strain-promoted azide–alkyne click chemistry (SPAAC). MAO-SiR exhibits exceptional photostability, significantly reducing photobleaching issues in extended imaging sessions. Its non-toxic nature ensures safe application across various cell types, including delicate primary cardiomyocytes. Furthermore, MAO-N3′s compatibility with sophisticated imaging techniques such as confocal, SIM, SMLM, and STED microscopy, facilitates detailed visualization of complex mitochondrial structures, like cristae, with superior spatial and temporal resolution. Note: In time-lapse imaging, cells treated with 200 nM MAO-N3 and 100 nM SiR-DBCO, collectively known as MAO-SiR, exhibit minimal signal degradation and a rise in cell density, indicative of robust mitochondrial function and cell health. This outcome is in stark contrast to the results observed with dyes like MitoTracker Deep Red, which typically show a substantial decline in signal intensity within just a few hours.
