General description
Serine/threonine-protein kinase Nek7 (UniProt: Q8TDX7; also known as EC:2.7.11.1, Never in mitosis A-related kinase 7, NimA-related protein kinase 7) is encoded by the NEK7 gene (Gene ID: 140609) in human. NEK7 is a protein kinase that plays an important role in mitotic cell cycle progression . It is highly expressed in lung, muscle, testis, brain, heart, liver, leukocyte, and spleen. Lower expression is observed in ovary, prostate, and kidney. It is required for microtubule nucleation activity of the centrosome, robust mitotic spindle formation, and cytokinesis. It is reported to phosphorylate echinoderm microtubule-associated protein-like 4 (EML4) at serine 146 and promotes its dissociation from microtubules during mitosis which is required for efficient chromosome congression. It acts as an essential activator of the NLRP3 inflammasome assembly independently of its kinase activity by unlocking NLRP3 following NLRP3 translocation into the microtubule organizing center, relieving NLRP3 autoinhibition and promoting formation of the NLRP3:PYCARD complex, and activation of CASP1. NEK7 is shown to be autoinhibited by the protrusion of tyrosine 97 into the active site and this inhibition is relieved by binding to the non-catalytic C-terminal domain (aa 810-828) of NEK9. Y97A, Y97L, or Y97F mutations in NEK7 results in more active kinase. Application
Quality Control Testing
Evaluated by Western Blotting in Human lung tissue lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected NEK7 in Human lung tissue lysate.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected NEK7 in Human skeletal muscle.
Immunocytochemistry Analysis: A 1:1000 dilution from a representative lot detected NEK7 in HeLa cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1C10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Serine/threonine-protein kinase Nek7 (NEK7). It targets an epitope within 16 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
