General description
Histone-lysine N-methyltransferase SETD5 (UniProt: Q9C0A6; also known as SET domain-containing protein 5, SETD5) is encoded by the SETD5 (also known as KIAA1757) gene (Gene ID: 55209) in human. SETD5 is shown to be essential for mammalian development and is required for regulating germ cell-associated genes in embryonic stem cells. It localizes to active transcribed genes and its SET domain is localized to amino acids 269-390. It interacts with Tbl1xr1 and Ctr9, which are components of 2 different transcriptional regulatory complexes (NcoR1 corepressor complex and Paf1 complex) in murine embryonic stem cells. SETD5-deficient embryonic stem cells display impaired cellular proliferation, increased apoptosis, defective cell cycle progression, a diminished ability to differentiate into cardiomyocytes, and greatly perturbed gene expression. SETD5-deficient mice embryos exhibit severe defects in neural tube formation, somitogenesis, and cardiac development, have aberrant vasculogenesis in embryos, yolk sacs, and placentas. They die between embryonic day 10.5 and 11.5. SETD5 may act by mediating trimethylation of lysine 36 of histone H3 (H3K36me3), which is essential to allow on-time RNA elongation dynamics. It can also monomethylates lysine 9 of histone H3 (H3K9me1) in vitro. Three isoforms of SETD5 have been described that are produced by alternative splicing. Mutation in SETD5 gene has been linked to intellectual disability. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell nuclear extract.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SETD5 in HeLa cell nuclear extract.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SETD5 in Raji cell nuclear extract.
Affinity Binding Assay: A representative lot of this antibody bound recombinant fragment of Human SETD5 with a KD of 9.7 x 10-9 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SETD5 in HeLa cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 3B22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects SET domain-containing protein 5 (SETD5). It targets an epitope within the internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
